Abstract

Polycyclic aromatic hydrocarbons (PAHs) are compounds with two or more condensed aromatic rings, that are hydrophobic, toxic, resistant to degradation, and pose a threat to human health. Microorganisms play a leading role in the transformation and utilization of PAHs in the environment. Phenanthrene-degrading strain 8G4 was isolated from soil in the Verkhnekamskoye salt deposit (Solikamsk, Perm Krai). Based on 16S rRNA gene analysis, the strain was identified as a member of the genus Stutzerimonas. The strain is capable of growth on naphthalene, salicylate, and benzoate as substrates, but does not grow on ortho-phthalate. This suggests that phenanthrene degradation by the strain is possible with the formation of 1-hydroxy-2-naphthoic acid, salicylate, and catechol as key metabolites. The strain was found to be capable of growth in a salt-free medium, as well as in the presence of up to 9% NaCl in a complete medium, and up to 7% NaCl in a mineral medium with phenanthrene as a substrate. The highest value of the maximum optical density of the culture, when grown on phenanthrene, was recorded in a medium containing 3% NaCl. An experiment with washed cells showed that strain 8G4 utilized 99.2% of phenanthrene in a medium without NaCl and 81.2% of it in a medium containing 7% NaCl, which has not previously been demonstrated for bacteria of the genus Stutzerimonas. Strain Stutzerimonas sp. 8G4 holds promise for further research and the development of new biotechnologies for the treatment of PAH-contaminated soils and industrial wastewater.
